Long-term segmentation-free assessment of head–flagellum movement and intracellular calcium in swimming human sperm

چکیده

ABSTRACT Human spermatozoa are the archetype of long-term self-organizing transport in nature and critical for reproductive success. They utilize coordinated head flagellar movements to swim long distances within female tract order find fertilize egg. However, date, analysis sperm head–flagellar movements, or indeed those other flagellated microorganisms, remains elusive due limitations microscopy flagellar-tracking techniques. Here, we present a novel methodology based on local orientation isotropy bio-images obtain kinematic physiological parameters individual free-swimming without requiring image segmentation (thresholding). This computer-assisted segmentation-free method evaluates, first time, characteristics movement beating up 9.2 min. We demonstrate its powerful use by showing how releasing Ca2+ from internal stores significantly alters behavior. The allows straightforward generalization bio-imaging applications, such as studies bull Trypanosoma, microorganisms – appealing communities than investigating biology.

Acetylcholine causes an increase of intracellular calcium in human sperm.

Sperm nicotinic acetylcholine receptors (nAChRs) can influence motility and the initiation of acrosome reaction (AR). We report that AR initiation by acetylcholine (ACh) in capacitated human sperm requires both Na+ and Ca2+ in the external medium. Pre-incubation with 50 microM 3-quinuclidinyl benzilate (QNB) or 50 nM strychnine failed to inhibit the ACh-initiated AR, demonstrating that muscarin...

Human recombinant interferon gamma enhances neonatal polymorphonuclear leukocyte activation and movement, and increases free intracellular calcium

In previous studies, we have reported that after chemotactic factor stimulation, PMNs from neonates fail to undergo certain critical activation steps. Furthermore, the concentration of free intracellular calcium reached is significantly below that of PMNs from adults. Interferon-gamma (IFN-gamma) is a lymphokine that has been shown to activate phagocytic cells, and IFN-gamma messenger RNA produ...

Inhibition of CatSper and Hv1 Channels and NOX5 Enzyme Affect Progesterone-Induced Increase of Intracellular Calcium Concentration and ROS Generation in Human Sperm

Background: Normal sperm function depends on appropriate intracellular calcium (Cai2+) and reactive oxygen species (ROS) levels. Calcium activates NADPH oxidase-5 (NOX5) that leads to ROS generation. The calcium channel of sperm (CatSper) is activated by progesterone and intracellular alkalization. Herein, the interactive role of CatSper, Hv1 channels, and NOX5 enzyme on Cai2+ and ROS generatio...

Introduction: Aspartame is the most famous and widely used artificial sweetener which is extensively used in food stuffs. Many controversial reports are presented on the toxisity of aspartame on different body tissues; nonetheles, little data is available about the the side effects of Aspartame on the reproductive system. The present study was conducted in order to evaluate the effects of aspar...
